Cheese whey level and caseinomacropeptide (CMP) index of fermented milk beverages added with four levels of cheese whey (0, 10, 20, and 40%) and stored at 8-10oC for 0, 7, 14 and 21 days were determined by high performance liquid chromatography-gel filtration (HPLC-GF). Additionally, the interference of the starter culture and the storage time on the detection of cheese whey and CMP were investigated. Refrigerated storage up to 21 days did not affect (P>0.05) cheese whey and CMP amounts in milk (0% of cheese whey) and in fermented milk beverages added with 10 and 20% of cheese whey (P>0.05). However, cheese whey and CMP amounts were higher than expected (P<0.05) in fermented milk beverages added with 40% of cheese whey and stored for 21 days. |

The aim of this study was to detect and quantify the main residual monomers released from composites, using high performance liquid chromatography (HPLC). Discs were made with dental composites (Herculite XRV, Tetric Ceram and Filtek Z250) and immersed in deionized water at 37ºC for 28 days, with water changes in 1, 7, 14 and 21 days. The mean concentration of residual monomers were subject to the Kruskal-Wallis test (p<0.05). Tetric Ceram exhibited significantly higher concentrations of leached monomers. Bis-GMA was the monomer released in lower concentrations for all the materials. There was no statistical difference between the amounts of TEGDMA and UDMA. Most of the monomers demonstrated maximal concentration at the 7-day period. The HPLC analysis... |

The current study evaluated antidiabetic and antihyperlipidemic activities of aqueous extract of Sigesbeckia orientalis L. (St. Paul’s Wort) (AESO) in an alloxan-induced diabetic rat model. Following OECD guidelines safe doses of AESO were assessed in rats for the main study. Serum/bood glucose, α-amylase, and lipids levels and histopathological evaluations were conducted to assess antidiabetic and associated antihyperlipidemic efficacies of AESO. AESO was found to be safe up to the dose of 2000 mg/kg. Significant (p < 0.05) reduction in glucose and lipids (total cholesterol, triglycerides, low-density lipoproteins) levels was observed in AESO treatment groups. Serum α-amylase, high-density lipoproteins, and total body weight was increased significantly... |

A HPLC protocol is described which permits analysis of the pigment composition of coastal and intertidal sediments. The HPLC technique has been extensively applied to phytoplankton pigments for fifteen years. Papers dealing with living benthic microphytic populations have been more rare, due to complexity existing in the sediment surface, where pigments (chlorophylls and carotenoids) from several taxonomic groups of microalgae are mixed with degraded pigments from senescent phytoplankton cells and macrophytic material. The present paper describes in detail a protocol adapted from Kraay et al. (1992), which allows a good discrimination of photosynthetic pigments of microphytobenthos communities. The following procedure is recommended. Sediment samples are... |

ABSTRACT Soybeans contain about 30% carbohydrate, mainly consisting of non-starch polysaccharides (NSP) and oligosaccharides. NSP are not hydrolyzed in the gastrointestinal tract of monogastric animals. These NSP negatively affect the development of these animals, especially the soluble fraction. This work aimed to establish a method to quantify NSP in soybeans, using high performance liquid chromatography (HPLC), and to estimate correlations between NSP, oligosaccharides, protein and oil. Sucrose, raffinose + stachyose, soluble and insoluble NSP contents were determined by HPLC. Oil and protein contents were determined by near-infrared spectroscopy (NIRS). The soluble PNAs content showed no significant correlation with protein, oil, sucrose and raffinose... |

Chloropigments and carotenoids were measured by HPLC in an intertidal muddy sediment of Marennes-Oleron Bay (France). Concentrations were determined as a function of sediment depth at low tide. The analyses were carried out at monthly intervals over 1 yr. Pigment analysis indicated that the microphytobenthic community was dominated by diatoms throughout the sampling year. Chlorophyll b was not encountered at any time or depth, indicating that no input from macrophytic detritus had occurred at the sampling site. There was a large pool of phaeopigments a, of which phaeophorbides were the major forms (75%). A microphytobenthic bloom occurred between March and June during which phaeopigments significantly increased with a high proportion of phaeophorbides... |

Abscisic acid (ABA) and indole-3-acetic acid (IAA) participate in the propagation of plants by somatic embryogenesis, causing polar structural differentiation of the embryo. The goal of the assay was to compare endogenous levels of ABA and IAA between somatic embryos (SE) and zygotic embryos (ZE) of Nothofagus alpina (Poepp. & Endl.) Oerst. In this study, a somatic embryo maturation assay involving the addition of varying concentrations of exogenous ABA was performed on cotyledonary-stage of N. alpina. Furthermore, the endogenous levels of ABA and IAA were quantified in the immature ZE, the mature ZE, and the embryonic axis of a mature embryo of N. alpina. The current study utilized high performance liquid chromatography (HPLC) for quantification.... |

In order to evaluate the influence of photoperiod on the accumulation of allantoin in comfrey (Symphytum officinale L.), a well known medicinal plant, an experiment was conducted during July and August, 1998. Cuttings obtained from 90 day old plants were submitted to four photoperiods (8, 12, 16 and 20 hours). After 60 days, allantoin content in roots and rhizomes was evaluated. The results showed that increases in photoperiod promoted an increment in the average content of allantoin in roots (0.06%, 0.303%, 1.213% and 4.78%). On the other hand, in rhizomes, allantoin accumulation decreased (9.65%, 7.14%, 0.55%) when the photoperiod was increased from 8 to 12 and 16 hours, respectively, stabilizing on a 20-hour photoperiod (0.53%). Plants cultivated under... |

Rivaroxaban, an anti-clotting medication, acts at a crucial point in the blood-clotting process and stops the formation of blood clots. In this study, RP-HPLC method was developed for the determination of rivaroxaban in tablets (Xarelto® (10 mg)). Phenomenex Luna 5 µm C18 100 Å LC Column (250 x 4.6 mm) was used at 40 ºC. Isocratic elution was performed with ACN:Water (55:45 v/v) mixture. The flow rate was 1.2 mL min-1 and UV detection was at 249 nm. Internal standard (Caffeine) and rivaroxaban were eluted within 2.21 and 3.37 minutes, respectively. The developed method was validated according to the ICH guidelines and found to be linear within the range 0.005 - 40.0 µg mL-1. The method was accurate, precise, robust and rapid. Thus, it was applied... |

In this work, polyclonal antibodies anti-human Factor IX were produced in New Zealand rabbits by immunization with commercial pure human FIX (hFIX) (Octanyne®, Octapharma, USA). The serum containing immunoglobulins anti-hFIX was useful to detect hFIX antigen in human plasma fractions submitted to anionic exchange chromatographic process and with a large yield. Immunoassays (ELISA) using bovine serum albumin, trypsin and peptides generated by cleavage assays with trypsin as digestion enzyme was performed and revealed adequate specificity of the polyclonal antibodies produced. |

Samples of phytoplankton cultures and of suspended particulate matter from continental, brackish and marine waters were examined by HPLC for the monosaccharide content of their hydrolysable sugar fraction. Eight monosaccharides were currently assayed and their weight compared to HPLC-assayed chlorophyll a concentrations. With the exception of oligotrophic waters, these two variables are well correlated in the various prospected environments. Glucose was always the strongly dominant form, not only in fresh and brackish waters but also in marine samples with high phytoplanktonic biomass. If samples are compared on a salinity basis, only four monosaccharides showed significant differences in their relative concentrations, as a consequence of sampling... |

The objective of this study was to evaluate the flow of nitrogenous compounds, protein degradability, rumen degradation of total carbohydrate and organic matter and microbial efficiency in heifers subjected to diets containing corn silage, sugarcane or Tifton. For this purpose, the 2,6-diaminopimelic acid (DAPA) technique was adopted and analytical procedures for amino acids by HPLC were adapted. Six rumen-fistulated Holstein-Zebu heifers with 480 kg of initial BW and at 24 months of age kept in individual tie stalls were assigned to two 3 × 3 Latin squares. Omasal digesta dry matter and microbial dry matter flows were determined using the isolated, purified and enriched lignin (LIPE®) and DAPA markers, respectively. Isolated bacteria from rumen showed on... |
