Distribuição de massa molecular de ácidos húmicos e promoção do crescimento radicular Rev. Bras. Ciênc. Solo

Detection of cheese whey and caseinomacropeptide in fermented milk beverages using high performance liquid chromatography Arq. Bras. Med. Vet. Zootec.
Andrade,E.H.P.; Souza,M.R.; Fonseca,L.M.; Penna,C.F.A.M.; Cerqueira,M.M.O.P.; Roza,T.; Seridan,B.; Resende,M.F.S.; Pinto,F.A.; Villanoeva,C.N.B.C.; Leite,M.O..
Cheese whey level and caseinomacropeptide (CMP) index of fermented milk beverages added with four levels of cheese whey (0, 10, 20, and 40%) and stored at 8-10oC for 0, 7, 14 and 21 days were determined by high performance liquid chromatography-gel filtration (HPLC-GF). Additionally, the interference of the starter culture and the storage time on the detection of cheese whey and CMP were investigated. Refrigerated storage up to 21 days did not affect (P>0.05) cheese whey and CMP amounts in milk (0% of cheese whey) and in fermented milk beverages added with 10 and 20% of cheese whey (P>0.05). However, cheese whey and CMP amounts were higher than expected (P<0.05) in fermented milk beverages added with 40% of cheese whey and stored for 21 days.

Growth, delayed fluorescence and pigment composition of four Prorocentrum minimum strains growing at two salinities Biol. Res.
BERDEN-ZRIMEC,MAJA; FLANDER-PUTRLE,VESNA; DRINOVEC,LUKA; ZRIMEC,ALEXIS; MONTI,MARINA.
Prorocentrum minimum is a potentially harmful and widely distributed marine dinoflagellate. Several P. minimum strains have already been studied, showing phylogenetical relations of strains isolated from the same geographical regions. Similarity among the strains was further examined on the basis of their physiology. Pigment composition and concentration, as well as delayed fluorescence (DF) decay kinetics and intensity, were measured in four P. minimum strains isolated from the Baltic and Adriatic Seas. The strains were grown at two salinities characteristic of the Baltic (8 PSU) and North Adriatic Seas (32 PSU). Strain differences in DF decay kinetics and growth did not always follow their genetic relations. While two strains showed similarities to the...

Identification et quantification des pigments chlorophylliens et caroténoïdes des sédiments marins : un protocole d'analyse par HPLC ArchiMer
Brotas, V; Plantecuny, Mr.
A HPLC protocol is described which permits analysis of the pigment composition of coastal and intertidal sediments. The HPLC technique has been extensively applied to phytoplankton pigments for fifteen years. Papers dealing with living benthic microphytic populations have been more rare, due to complexity existing in the sediment surface, where pigments (chlorophylls and carotenoids) from several taxonomic groups of microalgae are mixed with degraded pigments from senescent phytoplankton cells and macrophytic material. The present paper describes in detail a protocol adapted from Kraay et al. (1992), which allows a good discrimination of photosynthetic pigments of microphytobenthos communities. The following procedure is recommended. Sediment samples are...

Quantification of anti-nutritional factors and their correlations with protein and oil in soybeans Anais da ABC (AABC)
BUENO,RAFAEL D.; BORGES,LEANDRO L.; GOD,PEDRO I.V. GOOD; PIOVESAN,NEWTON D.; TEIXEIRA,ARLINDO I.; CRUZ,COSME DAMIÃO; BARROS,EVERALDO G. DE.
ABSTRACT Soybeans contain about 30% carbohydrate, mainly consisting of non-starch polysaccharides (NSP) and oligosaccharides. NSP are not hydrolyzed in the gastrointestinal tract of monogastric animals. These NSP negatively affect the development of these animals, especially the soluble fraction. This work aimed to establish a method to quantify NSP in soybeans, using high performance liquid chromatography (HPLC), and to estimate correlations between NSP, oligosaccharides, protein and oil. Sucrose, raffinose + stachyose, soluble and insoluble NSP contents were determined by HPLC. Oil and protein contents were determined by near-infrared spectroscopy (NIRS). The soluble PNAs content showed no significant correlation with protein, oil, sucrose and raffinose...

Monthly HPLC measurements of pigment concentration from an intertidal muddy sediment of Marennes-Oléron Bay, France ArchiMer
Cariou-le Gall, Valerie; Blanchard, Gerard.
Chloropigments and carotenoids were measured by HPLC in an intertidal muddy sediment of Marennes-Oleron Bay (France). Concentrations were determined as a function of sediment depth at low tide. The analyses were carried out at monthly intervals over 1 yr. Pigment analysis indicated that the microphytobenthic community was dominated by diatoms throughout the sampling year. Chlorophyll b was not encountered at any time or depth, indicating that no input from macrophytic detritus had occurred at the sampling site. There was a large pool of phaeopigments a, of which phaeophorbides were the major forms (75%). A microphytobenthic bloom occurred between March and June during which phaeopigments significantly increased with a high proportion of phaeophorbides...

Extraction time for soybean isoflavone determination BABT
Carrão-Panizzi,Mercedes C.; Favoni,Silvana Pedroso de Goés; Kikuchi,Akio.
Studies were carried out on the optimization of the extraction procedures for isoflavones determination in soybean by HPLC. Isoflavones were extracted from 100 mg samples of full fat soybean flour "Kinako" (milled toasted grains). Recovery of average total isoflavones was higher when extraction was performed with agitation(217.2 mg/100g), than without agitation(191.0 mg/100g). Isoflavone extraction without agitation for 1,4, 20 and 24 hours, were equally efficient for total and individual compounds. These results suggested that an efficient isoflavone recovery could be achieved with extraction for one hour with agitation.

Endogenous Quantification of Abscisic Acid and Indole-3-Acetic Acid in Somatic and Zigotic Embryos of Nothofagus alpina (Poepp. & Endl.) Oerst Chilean J. Agric. Res.
Cartes Riquelme,Pricila; Ríos Leal,Darcy; Sáez Carrillo,Kátia; Uribe Moraga,Matilde; Valenzuela Aguilar,Sofía; Joseph Bolus,Stephen; Sánchez Olate,Manuel.
Abscisic acid (ABA) and indole-3-acetic acid (IAA) participate in the propagation of plants by somatic embryogenesis, causing polar structural differentiation of the embryo. The goal of the assay was to compare endogenous levels of ABA and IAA between somatic embryos (SE) and zygotic embryos (ZE) of Nothofagus alpina (Poepp. & Endl.) Oerst. In this study, a somatic embryo maturation assay involving the addition of varying concentrations of exogenous ABA was performed on cotyledonary-stage of N. alpina. Furthermore, the endogenous levels of ABA and IAA were quantified in the immature ZE, the mature ZE, and the embryonic axis of a mature embryo of N. alpina. The current study utilized high performance liquid chromatography (HPLC) for quantification....

Influence of photoperiod on the accumulation of allantoin in comfrey plants Rev. Bras. Fisiol. Veg.
CASTRO,ANA HORTÊNCIA FONSÊCA; YOUNG,MARIA CLÁUDIA MARX; ALVARENGA,AMAURI ALVES DE; ALVES,JOSÉ DONIZETI.
In order to evaluate the influence of photoperiod on the accumulation of allantoin in comfrey (Symphytum officinale L.), a well known medicinal plant, an experiment was conducted during July and August, 1998. Cuttings obtained from 90 day old plants were submitted to four photoperiods (8, 12, 16 and 20 hours). After 60 days, allantoin content in roots and rhizomes was evaluated. The results showed that increases in photoperiod promoted an increment in the average content of allantoin in roots (0.06%, 0.303%, 1.213% and 4.78%). On the other hand, in rhizomes, allantoin accumulation decreased (9.65%, 7.14%, 0.55%) when the photoperiod was increased from 8 to 12 and 16 hours, respectively, stabilizing on a 20-hour photoperiod (0.53%). Plants cultivated under...

Monosaccharide composition of suspended organic particulate matter in relation to its origin ArchiMer
Compiano, Am; Romano, Jc; Delagiraudiere, I; Laborde, P.
Samples of phytoplankton cultures and of suspended particulate matter from continental, brackish and marine waters were examined by HPLC for the monosaccharide content of their hydrolysable sugar fraction. Eight monosaccharides were currently assayed and their weight compared to HPLC-assayed chlorophyll a concentrations. With the exception of oligotrophic waters, these two variables are well correlated in the various prospected environments. Glucose was always the strongly dominant form, not only in fresh and brackish waters but also in marine samples with high phytoplanktonic biomass. If samples are compared on a salinity basis, only four monosaccharides showed significant differences in their relative concentrations, as a consequence of sampling...
